Method development is part of how we work. From the community-standard GreenGate cloning system to interactive explorers of single-cell expression data in shoot and root meristems.

GreenGate modular cloning

GreenGate is a Golden Gate–based system for rapidly assembling plant transformation constructs. It uses a single type IIS restriction enzyme and just six classes of insert module — plant promoter, N‑terminal tag, coding sequence, C‑terminal tag, plant terminator and plant resistance cassette — and lets several expression cassettes be stacked into one binary destination vector from a library of pre-cloned building blocks. It is cheap, reliable, and now used by plant labs worldwide.

The original modular system for rapid, cheap and reliable assembly of plant transformation constructs. The complete set of GreenGate entry and destination vectors is distributed as a kit through Addgene, with maps, sequences and documentation for every module — request the plasmids directly for use in your own transformation pipeline.

Extension — GreenGate 2.0

The Maizel lab GreenGate 2.0 addons

An important, backwards-compatible extension developed by the neighbouring Maizel lab at COS Heidelberg. GreenGate 2.0 adds one-pot entry-clone generation (a Universal Entry Generator, pUEG), up to twelve Level 0 modules per transcriptional unit, the GreenBraid method for iterative stacking of multigene constructs, and ready-made modules for inducible and fluorescent markers — while remaining compatible with most existing GreenGate modules.

Single-cell expression explorers

Lab internal

To make our single-cell and single-nucleus transcriptome data explorable at the bench, we build interactive viewers that let you paint gene expression directly onto the tissue. These tools are currently provided for lab-internal use only and are protected by lab credentials.

3D SAM single-cell expression explorer

An interactive 3D viewer of a segmented Arabidopsis shoot apical meristem. Query any gene by symbol or AGI to paint its expression cell-by-cell across the reconstructed meristem, step through gene lists, and find genes with similar spatial patterns — with young and old primordia painted from their matched scRNA maturation stage. An interactive 2D viewer of the Arabidopsis root tip. Query any gene by symbol or AGI to paint its expression across the cell types of the root — from the stem cell niche and columella through the meristematic and elongation zones — directly from single-cell RNA-seq.
